Cell rolling is known as the leukocyte cell homing, which is represented by the continuous interaction between cellular surface receptor and expressed ligand on the endothelial cells. This mechanism strictly controls the rolling velocity of leucocytes toward the inflammatory region. In our previous work, the surface marker and its density were successfully converted to the rolling velocity of cells on ligand-immobilized column, and therefore the subpopulations of the cells were isolated on this column. This mechanism is applicable for evaluating the cell surface marker patterns under the labeled free conditions. In this study, cell-rolling microchip was developed for detecting the peripheral circulating cells. In recent study, it is revealed that cancer metastasis induced by the circulating tumor cells (CTCs). Primary diagnosis of early stage cancer disease should be the detection of CTCs. Copolymers of poly[2-methacryloyloxyethyl phosphorylcholine (MPC)-co-n-butyl methacrylate (nBMA)-co-N-vinylformamide (NVF)] were synthesized for the immobilization of the antibody as the ligand for cell rolling. Microflow pass surface (width: 300�m, depth: 150�m) was covered with the polymers, and atni-CD34 antibody were covalently conjugated through NVF segment. When we employed the optimized condition at 0.4 �l/min of media flow, the rolling velocity of KG-1a (CD 34 positive) cells was detected as about 40 �m/sec. On the other hands, the negative cells were shown the velocity as more than 55 �m/sec.  Significant difference between these rolling velocities was observed on the microchip system. From these results, we successfully discriminate the cell type on the microchip by the cell-rolling velocity without any non-specific absorption.
