Fabrication of triaxial biodegradable nanofibers by multi-axial electrospinning is receiving increasing attention in tissue engineering. However, due to the low contrast of electron densities between polymer phases and the overlapping of layers, the characterization of core/shell and triaxial structure is difficult. In this study, a biodegradable, multi-layer nanofiber structure has been successfully developed by triaxial electrospinning with gelatin as the sheath, poly(lactide-co-glycolide) (PLGA) as the middle layer and heparin as the core. Different techniques are applied to demonstrate the multi-layer structure , such as confocal fluorescence microscopy (CFM), focused ion beam and field emission scanning electron microscopy (FIB-FESEM), cross section transmission electron microscopy  (TEM) and nanoscale infrared spectroscopy (Nano-IR).  The results will not only enable the triaxial electrospinning technique to have wider applications in biotechnology, but also develop the characterization of core/shell and multi-layer electrospun structure.
