The anti-establishment complex, composed of the WAPL and PDS5 proteins in yeast and humans, normally serves to maintain sister chromatid cohesion by stabilizing the interaction between cohesin and the chromosomes. The WAPL/PDS5 complex also interacts with cohesin just prior to anaphase to remove it, allowing the chromosomes to separate and migrate to the cellular poles. The roles and functions of PDS5 homologues in Arabidopsis thaliana were investigated.Five putative PDS5 genes have been identified in Arabidopsis thaliana based on sequence homology; these have been re-named as PDS5-1 through PDS5-5. Homozygous knockout lines were generated for each of the genes, in addition to 'double' homozygous knockout lines for each possible pair of genes. Transgenic lines were confirmed via PCR, and were then subjected to general phenotypic analysis. Results show that the genes play roles in vegetative growth, flowering, and seed development, and that at least two of the genes functionally compensate for one another.
