Changes in expression of regulatory RNAs have been implicated in various diseases. The purpose of this study is to test how the expression of several miRNAs in human lung cells help respond to stress from exposure to various environmental chemicals. The specific group of miRNAs involved in this study has been identified in mouse lung irritation studies. We will present results collected after exposing the model A549 lung cells to ozone and various carbonyl mixtures. Exposures under these conditions were precisely controlled in specially constructed environmental chambers. We have designed several specific probes and monitored the miRNA expression patterns by Quantitative-RT PCR. Our results suggest that miRNAs could globally regulate cellular responses to environmental changes. Given the known roles for miRNAs in developmental regulation, these findings demonstrate a significant new link between air pollutants and human health at the molecular level.
