The 2D IR approach has been utilized for direct studies of conformational dynamics of peptides. Recently, temporal evolution of local structural changes was observed during helix reorganization from a distribution near, but not initially at the equilibrium distribution [1]. Understanding specific structural changes associated with protein folding and misfolding provide insights into the molecular movements leading to protein function. Well-chosen infrared markers can provide the ability to obtain site-specific structural changes of biomolecules while simultaneously detecting the local dynamics in the region where biological function takes place. The coupling between vibrational modes as well as the detection of frequency fluctuations resulting from the local environment is then tracked by 2D IR. [1] PNAS 2013; doi:10.1073/pnas.1311876110
