Batrachotoxin, a complex steroidal alkaloid, was first isolated from the Colombian poison arrow frog, Phyllobates aurotaenia. The structure features a steroid backbone functionalized with a transannular hemiketal and fused oxazapane ring. Biological studies have revealed it as the most toxic non-peptidal natural product known, with its potency resulting from strong agonist activity towards voltage-gated sodium ion channels. Our interest in the structure, synthesis, and biological activity of batrachotoxin has led us to pursue its total synthesis. A summary of the current efforts towards the total synthesis will be presented.
