Recent studies have suggested that hydroxylated metabolites of polybrominated diphenyl ethers (OH-PBDEs) accumulate in human serum at levels similar to or greater than their parent PBDEs, and have the potential to cause endocrine disruption. Conventionally, gas chromatography/mass spectrometry (GC/MS) after derivatization is used to analyze for OH-PBDEs. However, there is a need for the direct analysis of OH-PBDEs at relatively low concentrations in environmental and biological samples. Therefore, we explored the use of supercritical fluid-based convergence chromatography coupled to time of flight mass spectrometry method towards a non-target analysis to identify metabolites formed during in vitro metabolism of PBDE-100 by a recombinant human cytochrome P450 (CYP2B6). Without prior derivatization, five monohydroxylated metabolites were effectively separated by convergence chromatography, with the accurate masses of fragment ions containing bromine isotopic patterns clearly observed. In addition, convergence chromatography/mass spectrometry proved effective in the targeted analysis of OH-PBDEs in human serum samples.
