Natural products total synthesis provides a rich and unparalleledopportunity to develop new synthetic transformations, conceive novel andgeneral strategies to access complex structures, and study the mechanismof action of bioactive targets.  We have developed a general andversatile route to prepare complex diazofluorenes, an unusual functionalgroup found in the antiproliferative metabolites known as the kinamycinsand lomaiviticins.  Applications of this chemistry towards the synthesesof kinamycin F and lomaiviticin aglycon, and elucidation of the mechanismof action of lomaiviticin A, will be presented.
