4-Dimethylaminopyridine (DMAP) was found to have general applicability for the catalysis of a wide variety of reactions, especially in acylation reactions. A new series of chiral DMAP catalysts that incorporate fluxional groups have been designed. These catalysts were found to be efficient in promoting acylative kinetic resolution of sec-alcohols and axially chiral biaryls with selectivity factor up to 35 and 51 respectively. These results and application of the new DMAP catalysts in other transformations will be discussed.
