DNA lesions, caused by environmental agents as well as metabolic byproducts, can interfere with replication and transcription process. RNA polymerase has been proposed to be a highly selective DNA damage sensor, as it constantly scans the transcribed genome.RNA polymerase II (pol II) can stall or bypass some types of bulky DNA lesions. However, the molecular basis of how pol II recognizes and processes these DNA lesions is not fully understood. Here we used a combined structural and biochemical approach to gain novel insights in understanding the impact of these bulky DNA lesions on pol II transcriptional fidelity and how Pol II recognizes and processes bulky DNA lesions.
