Libraries of ferrocenes with defined redox properties are important in many areas of electrochemistry. In particular, their application in the medical diagnostics industry is an ever expanding area. Of particular interest has been the development of ferrocene-based probes for use in electrochemical sensing methodologies to allow for the detection of multiple specific gene fragments.This research has focussed on the development of a ferrocene library with a wide range of oxidation potentials, with the goal of developing five complimentary ferrocene probes that can be utilised in a �multiplex� fashion when utilised to detect multiple specific gene fragments from a PCR based assay.This goal has been achieved via two complementary processes: i) Major redox shifts have been achieved via incorporation of various organic functional groups onto the ferrocene core ii) �Environmental� fine tuning of the ferrocene oxidation potential has been achieved via modulating the hydrophobicity of the ferrocene through changes to the linker used to attach it to the probe. Ferrocenes produced viathese techniques have been utilised in a single test to detect multiple target analytes. On bench testing of these labels has shown clear differentiation of each of the five peaks in combination with a low background (Figure 1), in addition these probes have shown positive detection from only 10 copies of target DNA.
