Two water-soluble fluorescent probes based on BODIPY dyes bearing piperazine moieties have been developed for detection of intracellular pH (pHi). One probe responds to pHi through photo introduced electron transfer (PET) and the other is based on intramolecular charge transfer (ICT) mechanism which gives ratiometric fluorescence responses in near infrared region. The sensitivity and selectivity of these probes to pH have been investigated by spectroscopic analysis in aqueous solutions. Both of them have low fluorescence background at physiological neutral condition while their fluorescence intensities will dramatically increase with changes of pH from 7.4 to 4.0. Furthermore, these probes were successfully applied to the in-vitro imaging and monitoring of lysosome pH inside normal endothelial and breast cancer cells.
