A method based on liquid-liquid extraction and high performance liquid chromatography (HPLC) had been optimized for the determination of 6 polybrominated diphenyl ethers (PBDEs) congeners (BDE-1, BDE-7, BDE-8, BDE-17, BDE-18 and BDE-47). The factors relevant to the liquid-liquid extraction efficiency, such as the kind of extractant, the extraction time and the extraction times were optimized. The optimal conditions were as follows: dichloromethane as extractant, five min extraction time and extracted three times. The recovery rate was 98-102%. To quantify and completely separate the complex congeners in chromatography system, a response surface method (RSM) was employed to optimize the overall separation factors, which include mobile phase composition, flow rate and column temperature. The results indicate that the optimum levels of three variables were 96:4 of methanol/water volume ratio, 25 �C of column temperature and 1.2 ml/min flow rate. At the optimal condition, the method showed good linearity ( r > 0.996 over a concentration range 0.520 mg/l) and repeatability (relative standard deviation (RSD) < 2.5 % ). Furthermore, the detection limit (S/N = 3) of the method was 0.2 mg/L.
