Reaction-based ratiometric fluorescent probes have recently gained tremendous popularity in sensing applications because they allow quantitative measurements of analyte concentrations independent of the probe concentration. However, a pitfall of the majority of previously reported ratiometric probe designs is based on irreversible reactions.  In this talk, we will explain the importance and necessity of using reversible reactions for ratiometric probe design in order to quantitatively and reliably measure the analyte concentrations in biological systems. Additionally, we will demonstrate how a reversible reaction based ratiometric probe can quantitatively measure the intracellular glutathione concentration.
