Fastigiatine was isolated from the Lycopodium fastigatum in 1986 by Lock and co-workers. Its core structure contains a pentacyclic ring-system with five contiguous stereocenters, two of which are vicinal quaternary centers. Recent studies by the Kobayashi group led to the isolation of two new structures, Himeradine A and Lyconadine D, which contained the fastigiatine-type scaffold. The unique framework, as well as its biological activity, has drawn our attention to pursue the total synthesis of fastigiatine. Our synthetic approach towards fastigiatine will be presented.
