In this study, we demonstrate, for the first time, the dynamic introduction of bioactive RGD onto matrix via reversible multi-covalent interactions within phenylboronic acid (PBA)/cis-diol polymeric complexes.  In our design, bioactive RGD conjugated with a long glycopolymer chain was dynamically coupled on a matrix grafted with PBA polymer brushes, permitting dynamic and �reversible� control of cell adhesive properties.  Firstly, this new approach combines the advantages of both covalent and noncovalent interactions. The reversible multi-covalent interactions are more stable than noncovalent interactions, and the system also possesses dynamic properties because of its reversibility. Secondly, triggered-release of the coupled RGD can be achieved by specific biomolecular exchange, thus leading to biomolecule-regulated cell behavior. This system is different from previously reported biointerfaces which use external stimuli (e.g., UV light, temperature, potential, pH, etc.). Thus, this biomolecule-triggered approach is minimally invasive, and more importantly, it also provides opportunities for the mimic of natural biomolecular feedback systems. Also, �reversible� adhesion of the released cells in our system is also available. This is mainly because the accessibility of surface-conjugated long glycopolymer chains and their multiple stable interaction sites, which facilitate the process of molecular exchange and subsequent recruitment by the biointerfaces.In addition, the reversible multi-covalent interactions in PBA/cis-diol polymeric complexes in this work also provide a promising method for stable, dynamic immobilization of some relatively bulky biomolecules, such as DNA, RNA, and proteins, on artificial ECMs. Thus our system has more promising potential applications in the fields of medical diagnostics and regenerative medicine.
