Synthetic peptides have high demand in academic labs and industry. Some therapeutic peptides are on the market and several are in clinical trials. Peptide synthesis is carried out on automated solid phase synthesizers by stepwise addition of amino acid monomers. Besides full- length sequence, each step generates failure sequences and small molecules. The small molecules are generated from the side chain protecting groups. These small molecules are usually removed by cold diethyl ether precipitation. The failure sequences are difficult to remove because of similar physical properties to those of full length sequence. The known methods are expensive, needs large volume of solvent and time consuming for large scale purification. In this method, a polymerizable methacrylamide tag is attached to the desired full-length sequences, which are then incorporated into a polymer. Most of the impurities are removed by washing and the pure product is cleaved from polymer. This method does not need chromatography and expensive equipments and the purification is achieved by simple operations such as shaking, washing and extraction. The purified peptides have good purity and high recovery yield. Therefore, it is suitable for large scale purification of peptides.
