Stereospecific nickel-catalyzed cross-coupling methods were utilized to generate enantioenriched compounds for biological testing. 1,1-Diarylalkanes were synthesized through cross-coupling reactions of alkyl Grignard reagents, while 3,5-diaryl alcohol analogues were formed by nickel-catalyzed ring-opening reactions of cyclic ethers. A library of diarylalkanes and diaryl alcohols prepared by these methods was evaluated for anti-cancer activity against a range of cancer cell lines. Several compounds were found to selectively inhibit cancer cell proliferation in the low micromolar range.
