Within the area of sensing, signal amplification is essential for the detection of low concentrations of analytes. Over the past few decades, a multitude of amplification methodologies have emerged for the detection of a variety of analytes with low limits of detection (LOD), such as the polymerase chain reaction (PCR) and the dendritic chain reaction (DCR). Despite this, the need for fast and effective signal amplification strategies for the detection of analytes remains high, especially for rapid point-of-care testing (POCT). In this paper, we present the development of an enzyme-triggered catalytic signal amplification protocol. Figure 1. <img src='https://acs-archive-assets2.s3.amazonaws.com/241nm/uploads/74026_axyln60.jpg' alt='' />By utilizing a traditional enzyme-linked immunosorbent assay (ELISA) format combined with a metal-catalyzed transfer hydrogenation reaction, a hybrid (biological and synthetic) double amplification detection system is achieved, leading to both high selectivity and sensitivity. The enzyme-triggered catalytic signal amplification protocol is dependent upon a self-immolative pro-ligand; a synthetic compound inspired by trigger-linker-signal sensing methods. Once triggered by an enzyme, the pro-ligand releases a diamine ligand creating an active iridium catalyst for a transfer hydrogenation reaction. The active catalyst is then able to convert substrate to product, both of which are distinguishable electrochemically. The enzyme-triggered catalytic signal amplification protocol has been successfully applied to model analytes and is able to generate a positive response within minutes.
