Occurrences of pharmaceuticals and metabolites are evident in environmental samples and organisms. In this study, a reproducible gas chromatography - mass spectrometry (GC-MS) method is developed to determine anti-histamine diphenhydramine (DPH), anti-anxiety diazepam (DZP), anti-seizure carbamazepine (CZP) drugs and metabolites in environmental fish. Frequently detected 9-drugs and metabolites belonging from 3-classes of applications were analyzed by GC-MS using selected ion monitoring (SIM) from 14-edible fish fillets. Salt- and fresh-water grown fish collected from grocery stores were homogenized, extracted, pre-concentrated, cleaned up, and examined. Identifications of target compounds in extracts were established by comparing similar mass spectral features and retention properties with standards. Internal standard based calibration plots were used for quantitation. Of 9-target pharmaceuticals, DPH and DZP were observed with concentrations ranging from 0.61 to 6.21 and 1.99 to 16.57 ng/g, respectively. These values are about 2 to 10 times lower than those of environmental fish. Mean spike recoveries of target analytes exceeded over 75% with relative standard deviation (RSD) &lt; 10%. The statistically-derived method detection limits (MDLs) ranging from 0.13 to 5.56 ng/g could be achieved.
