In this work, we used single-molecule imaging to study the molecular-scale mechanisms of protein ion-exchange adsorption. We applied a combination of super-resolution optical methods and stochastic theory to obtain single-molecule adsorption and desorption kinetics of proteins on a realistic, agarose stationary phase. We have conclusively demonstrated that multiple-charge clusters are necessary for localized protein adsorption. Protein adsorption on dispersed singly-charged ligands was unobservable under our experimental conditions, with functional adsorption sites emerging at higher ligand surface loadings. We now are employing molecular-scale and ensemble approaches to investigate the competitive ion-exchange adsorption of proteins.
