In this study, we present a fast and convenient approach for developing a perfusion medium for antibody-producing Chinese hamster ovary (CHO) cells from a commercially available fed-batch medium and feeds (ActiCHO™ Media System). Applying a design of experiment (DoE) strategy, ActiCHO P base medium was used as a starting point and ActiCHO Feed-A and Feed-B were added in various concentrations as supplements. The DoE strategy consisted of both a screening and optimization design and the resulting medium prototypes were evaluated in batch cultures. Peak cell densities and antibody productivities under batch conditions using the best performing medium prototype in this study were more than doubled compared with the ActiCHO P base medium. In the next step, the best performing prototype was evaluated in small-scale perfusion cultures in rocking, single-use ReadyToProcess WAVE™ 25 bioreactor systems and the cell-specific perfusion rate (CSPR) was optimized. The best prototype medium in this study enabled perfusion culturing with a CSPR of &lt; 50 pL/cell/d.
