Mechanistic modeling for cation-exchange chromatography of monoclonal antibodies has been previously performed using the General Rate Model to describe transport behavior and the Steric Mass Action (SMA) model to describe protein binding. This presentation will explore a case study where the SMA model was not adequate to describe protein binding under high loading conditions for a monoclonal antibody. An alternative multi-state binding model will be proposed and utilized to successfully characterize protein binding behavior over the full range of loading conditions and potential mechanistic insight gained from the study will be discussed.
