Affinity chromatography is a powerful capture step for antibody and antibody fragment isolation from cell culture harvest due to its high specificity. In traditional batch chromatography, the capacity and productivity of the affinity stationary phases cannot be fully utilized due to mass transfer limitations, leading to sub-optimal relative resin costs and throughput.<br/>Due to sequential loading of two or more columns in series, counter-current cyclic capture processes combine high throughput with high loading capacity, allowing the maximal utilization of the stationary phase at reduced buffer consumption and therefore significant improvement of the process economics.<br/>In this talk the topics of process control and feed-back driven optimization strategies for counter-current cyclic capture processes are discussed. An online UV-based process tool capable of running the two strategies is presented in a case study for a twin-column counter-current capture process.
