Herein we report the synthesis and characterization of various analogs of biodegradable citrate elastomers with the ability to release nitric oxide (NO) for therapeutic purposes. Citrate elastomers are a naturally occurring, integral part of the bone nanocomposite. NO-releasing citrate elastomers show promising applications for preparing orthopedic scaffolds with improved bone regeneration capabilities, in cardiac tissue engineering, and drug delivery due to their enhanced mechanical properties. The elastomers were prepared by poly-condensation of maleic acid, citric acid, and 1,8-octanediol at 140°C for 2 hours. Incorporation of carboxylic acid-containing thiols preceded the polymerization in order to provide a reaction site for NO-donors. The thiolated complexes were then S-nitrosated by reaction with tert-butyl nitrite. NO-releasing materials are of interest due to the role of NO in physiological processes, such as regulation of vasodilation, modulation of inflammatory responses, and promotion of cell proliferation and angiogenesis. Thus, the extent of NO loading and release kinetics using real-time chemiluminescence-based NO analyzers were evaluated and related to the structure and environment associated with the thiol complex incorporated to the polymeric material. Degradation studies under physiological conditions (pH 7.4 phosphate buffer saline, 37 °C) were performed for 4 weeks, time in which the weight of polymer complexes decreased by 50%, confirming biodegradability.
