The novel bioanalytical methodology presented here is a potential clinical diagnostic tool, which allows for an isolation and examination of the size distribution of the metabolic variants of the triglyceride rich lipoprotein (TRL) subclass in human blood. The size of these particles may be an atherogenic risk factor, and it is important to develop technology to measure the varying size distributions of this subclass. Triglyceride rich lipoproteins (TRLs) are responsible for transporting dietary fats to cells throughout the body. Metabolically, TRL particles exist as a size distribution continuum dependent on triglyceride content as they are continuously metabolized by depositing fats to cells throughout the body. The main focus of our research is to develop methodology to easily isolate the TRL particles from blood plasma and to then characterize them via dynamic light scattering (DLS) analysis. We utilize an adapted sequential double spin high-speed centrifugal flotation technique to isolate the TRL subclass. Following this preparative isolation, DLS provides a quantitative size distribution of these particles. This methodology is a facile diagnostic tool that may be used in the clinical setting. While most clinics mainly assess LDL cholesterol, HDL cholesterol, and total triglycerides, it would be prudent to utilize these measurements as an additional tool in the assessment of atherogenic risk factors.
