Propinonibacterium acnes is best known as the cause of acnes vulgaris. In addititon, P. acnes causes post-operative and device-related infections, sarcoidosis and synovitis, pustulosis, hyperostasis, and osteitis. P. acnes is an anaerobic bacteria that is resistant to phagocytosis. Several P.acnes virulence factors have been identified, but few have been isolated and characterized. This work describes isolation and characterization of a P. acnes membrane-associated esterase. P. acnes was grown, and the cells were collected and lysed. Cell components were separated using differential centrifugation, and the fractions were assayed for esterase activity. Esterase activity was found in the cytoplasmic and membrane fractions. Membrane esterase activity was further isolated using gel filtration chromatography. Fractions containing esterase activity were pooled and studied using esterase inhibitors.
