In the course of optimizing  molecular interactions it is believed that while affinity is important, having enthalpy-based interactions can be particularly attractive.  It has been a long standing challenge to identify entalpic interactions due to the lack of correlation with molecular properties or absolute affinity.  The circumstances under which lipophilic efficiency (LipE) might be used to approximate changes in enthalpy of binding will be discussed.
