Measurement of protein concentration, especially at high concentration, is labor intensive which usually involves pausing UFDF operation, pulling representative samples, performing multiple gravimetrical dilutions if needed and then taking UV absorbance measurements in triplicate.  An online concentration measurement, if implemented for UFDF operation, will not only reduce labor intensive operations, but also providing real-time concentration information for process monitoring.<br/> <br/>This presentation explores Raman spectroscopic technology for online measurement of high protein concentration because of its following advantages: 1) it can provide specific information of analytes for both qualitative and quantitative analyses, 2) its nondestructive nature makes it an ideal process analytical tool for real time monitoring.<br/>Feasibility study covered in this talk uses a Kaiser in-process Raman probe with 785 nm laser excitation and 1 minute signal acquisition. Based on the protein specific Raman signal region, preliminary data suggests that Raman signal has a linear response within a wide mAb concentration range and with a dynamic buffer composition.   The results demonstrate that Raman spectroscopy probe is a feasible technology for online measurement of high protein concentration during UFDF.
