The monosaccharide <i>N</i>-acetyl-glucosamine (O-GlcNAc) can be posttranslationally added to proteins in the cytosol, nucleus, and mitochondria of cells. O-GlcNAcylation is required for development in mammals and contributes to a variety of human diseases. Here I will present the development and application of new metabolic chemical reporters (MCRs) of O-GlcNAcylation. Specifically, I will highlight the use of these molecules to both uncover unexpected cellular metabolism and identify key proteins in cell survival.
