The intriguing structural features, complexity, and biological activities of the phomoidrides have led to considerable interest from synthetic chemists. In our efforts toward a synthesis of phomoidride D, a synthetic strategy has evolved wherein a phenolic oxidation/intramolecular Diels-Alder sequence is followed by a SmI<sub>2</sub> mediated radical cyclization cascade to deliver a highly functionalized isotwistane intermediate that is poised to deliver the carbocyclic core via a late-stage Wharton fragmentation. These efforts along with studies directed toward completing the total synthesis of phomoidride D will be described.
