Inspired by Rosenberg's discovery of cisplatin, Escherichia coli is an informative model system to screen Ru(II) polypyridyl complexes with potential anti-cancer properties. E. coli also provides a simpler biological system to facilitate studies of mechanisms of action. Using the photo convertible protein Dendra2 as a reporter in E.coli, fluorescence-based assays are used to study the mechanisms of action of different Ru(II) polypyridyl complexes with potential anti-cancer activity. These studies were performed from the single cell level to population level. The kinetics of fluorescent protein production was used to evaluate the difference between agents that inhibit transcription vs. agents that act as inhibitors of translation. This research provides a simpler model system for biological studies to elucidate mechanistic details of cytotoxic agents.
