Increasing competitive and economic pressures are now driving the chemical and bio-manufacturing industries to implement process technologies that require less energy, generate less waste, require lower consumptive use of materials and consumables and employ lower capital investment. At the same time, more emphasis is being placed on product quality without sacrificing process economics in order to address public demands for greater product safety and efficacy. The need for continuous developments within the field of down stream technologies for biopharmaceuticals is symptomatic of the challenges now faced by researchers and manufacturers alike, although other fields of biomanufacturing, including the production of food ingredients and industrial enzymes, are equally dependent on new discoveries based on innovations in the future. In this context, a log jam has been evident for many years in biomanufacturing, due to the inadequacy of many down-stream processing procedures and their interfaced process analytical technologies to simultaneously, comprehensively and rapidly address issues of scale and productivity whilst achieving waste minimisation, cost efficiencies and product safety. How many of these key considerations will be resolved remain to be fully elucidated.<br/><br/>Central to these important issues is the question: “What challenges are already evident if the recovery of biological products is to be based on even more efficient and sustainable technologies?” Availability of increasingly powerful characterisation and process analytical technologies has opened up new avenues to address this question and provide insightful solutions. In this presentation, the linkages between the more routine adoption of these technologies and process supply chain considerations will be explored with a focus on the production of mid to higher value proteins based on affinity separation methods, as batch or semi-continuous procedures. In addition, several examples from the recent developments from this and other centres will be described, illustrating how method refinements or new approaches can be implemented to address more efficient, and in terms of resource utilisation, more sustainable production through the translation of basic science into innovations in the recovery of biological products.
