An emerging, vast target space for peptide drug discovery provides extraordinary opportunities for the development of novel breakthrough medicines and research tools to explore intracellular disease mechanisms involving protein-protein interactions. Macrocyclic peptides of varying classes of scaffolds are being advanced to modulate such intracellular PPI targets, including transcription factors and apoptotic regulatory proteins. This presentation will highlight intracellular targets and peptide design engaging innovative technologies that leverage structural biology, computational chemistry and molecular biophysics to re-define druggability.
