Glycoside hydrolases such as galactosidase are named based on their substrate specificity. However, usually the name only identifies one side of the two molecular components formed upon hydrolysis of the glycosidic linkage. Here, new mass spectrometry-based methods are reported that allow the complete substrate characterization of glycoside hydrolases, including thermostable versions, without the need for <i>p</i>-nitrophenol or other very reactive substrates. Unlabeled potential carbohydrate substrates can also be ordered in terms of their kinetic competency to give clues as to the likely natural substrate for a given enzyme.
