Calpain is a conserved family of calcium-dependent, cytosolic, neutral cysteine proteases. The best characterized members of the family are the ubiquitously expressed calpain 1 and calpain 2. Calpain are implicated in many physiological and pathological processes. They have significant role in the cell proliferation, differentiation and migration in a variety of mammalian cell types, contributing to the development of angiogenesis, vascular remodeling, and cancer. In an effort to investigate the effects of a newly synthesized calpain inhibitor (i.e., KR-185) on cell growth and differentiation we have performed preliminary studies to elucidate the effect of the inhibitor on the antioxidant status of human lung cancer cell line (A549) and 3T3-L1 preadipocytes respectively. The effects of the exposure of A549 cells and 3T3-L1 preadipocytes to several doses of KR-185 (at 0, 5 10, 15, 20 and 25 µM doses) for 24 hr on the expression levels of glutathione (GSH) (a natural antioxidant in most cells) were investigated. The results indicate that KR-185 did not have any effect on the GSH levels in both cell lines. GSH levels in both cell lines were higher for the treated groups compared to their respective controls. The trend in GSH expressions were similar for both cell lines for the doses of KR-185 tested except for the 15 µM where a slight decrease in GSH were observed. Thus, KR-185 maintians GSH in those cells and may indicate the ability of KR-185 to help those cells cope with oxidative damage especially in 3T3-L1 preadipocytes.
