We will describe the design, synthesis, and use of small molecule reagents for triaging samples in point-of-need assays. The presentation will describe readout reagent 1 as well as improved reagent 2. Mechanistic studies were used to determine the fate of thiol released. Ultimately, reagents 1 and 2 provide both qualitative and quantitative readouts in assays that detect enzyme biomarkers. The qualitative readout enables rapid on-site diagnosing of samples so that only samples containing relevant concentrations of the target analyte must be quantified.
