Development of synthetic methods to construct -OCF3 and -SCF3 is an active area of research as these moieties offer to provide desirable pharmacokinetic properties of the biologically active molecules. For example, higher lipophilicity and membrane permeability of potential drug molecules compared to -CF3 or -OCH3 analogs. Moreover, decarboxylative fluorination approach to build -OCF3 and –SCF3 offers opportunities in PET imaging studies. This work explores silver catalyzed decarboxylative fluorination of RXCF2CO2H (R = alkyl, aryl; X = O, S) to synthesize RXCF3 in the presence of an electrophilic fluorine source at ambient temperatures and shorter reaction times.
