Alzheimer’s disease is a neurodegenerative disorder characterized by the accumulation of neurotoxic peptide amyloid β 42 (Aβ<sub>42</sub>) in the brain. Currently 6 to 7 million people are afflicted with this devastating disease and USFDA approved drugs only provide temporary relief for six to 12 months. One of the approved drug tacrine has severe hepatotoxicity and has been discontinued. We recently studied the anti-glycation and anticancer properties of curcumin longa and okra seed extract (OSE) alone and also in combination with the well-established drug metformin (ACS Book Symposium Series 2012, ACS 2015). The combined effect of curcumin with metformin, and metformin with okra seed extract was found to be highly efficacious in inhibiting advanced glycation end-products (AGEs) which cause microvascular and macrovascular complications in diabetes (ACS AGFD 2014, 2015 ). In continuation of our studies the present research describes chemically engineered flavonoids present in okra seed extract by reacting methanolic solution of okra seed extract with hydrazine monohydrate in the domestic microwave oven for 50 seconds.The reaction in the microwave was stopped every 4 seconds interval and then continued for 45-50 seconds. The solvent methanol was evaporated under nitrogen and the resulting reaction mixture containing 3,5-diaryl pyrazole derivatives was resolved and analyzed via analytical and preparatory thin-layer chromatography. The structural characterization was accomplished by a combination of UV-visible NanoDrop spectrophotometry (OSE λ max. 358 nm vs. ammonolysis product λ max. 262 and 400 nm) and liquid-chromatography mass spectrometry (LC/MS/MS). These studies will be helpful not only in elucidating the mechanism of anti-glycation and anti-cancer activities but also binding of Aβ-oligomers and disaggregating fibrillar formation in Alzheimer’s disease. We believe that chemically engineered okra seed extract flavonoids provide another source of natural products which may be useful for studying nanoparticle drug delivery therapeutics as well.
