The expression of the cyclin-dependent kinase inhibitor p27Kip1 is reduced in many types of human cancer and inversely proportional to tumour aggressiveness. Indeed, p27KIP1 directly contributes to the regulation of cell cycle entry and withdrawl. Given the role of E2F1 in the activation of p27Kip1 expression, we prepared and tested several E2F-responsive luciferase reporters in a variety of cancer cell lines in order to identify modulators of E2F1. Identified modulators were then used to investigate the downstream signalling events involved in p27Kip1 mediated cell cycle control in cancer cells.
