The discovery of a synthetic ligand for a given protein of interest often begins with a high throughput screen of a large, chemical compound collection. This method of lead identification is costly, cumbersome, and frequently does not yield any significant leads from within the given chemical collection. Rapid and facile methods for screening even larger collections of more diverse molecules are needed. The proposed platform should enable the creation and evaluation of billion-member synthetic molecule collections, all within the space of a 96-well plate. Here we present progress towards the development of this new small molecule screening platform: 1) encoded library synthesis on sub-micron particles, 2) spatial arraying and en masse decoding in a microarray format, and 3) use of the microarray to identify ligands for a series of model proteins.
