The complexity of future armed conflict is partly due to the propagation of chemical, biological, radiological, nuclear, and explosive (CBRNE) threats and will require specially equipped forces to effectively work in inhospitable conditions. As ubiquitous and wearable sensor technology becomes the standard method of biosurveillance, the biorecognition elements integrated into such devices must be ruggedized for consistent performance outside of the laboratory and be capable of “on demand” production to quickly address emerging threats. The peptide-based Protein Catalyzed Capture (PCC) agent technology is a promising methodology to produce highly tailorable and highly stable biorecognition reagents through iterative <i>in situ</i> “click” chemistry cycles with one-bead-one-compound (OBOC) peptide libraries. Through this method, a linear biligand reagent was developed against a high priority Army target, the L1R protein of Vaccinia. More recently, our next generation cyclic peptide library has produced a PCC agent against L1R with ~2x better binding affinity in half the number of screening steps compared to the reagent produced utilizing our previous linear library. Since the reagent was discovered against an epitope away from the antibody immunogenic site on the protein through established epitope targeting capabilities, this PCC reagent can also be used as a reagent pair for sandwich assay applications. The current advances in screening methods afford the production of peptide-based antibody replacements with tailorable properties ideal for fieldable detection and monitoring platforms.
