In a similar fashion to the difficulty of forming quaternary carbon centers, alpha-tetrasubstituted amines remain a serious synthetic challenge. Few methods exist to convert commercial starting materials directly into the hindered amine scaffolds that form the core of numerous natural products and drug therapies. The development and application of catalyst systems capable of accessing and converting ketimine intermediates into tetrasubstituted building blocks will be discussed.
