A rational drug design, synthesis and anticancer activity evaluation of quinoline derived trifluoromethyl alcohols are described. This novel class of trifluoromethyl alcohols has also been identified as potent growth inhibitors in zebrafish embryo. The synthesis was achieved using a Csp3-H functionalization strategy of methyl quinolines with trifluoromethyl ketones. Furthermore, we explored the toxicity of ethyl 4, 4, 4-trifluoro-3-hydroxy-3-(quinolin-2-ylmethyl)-butanoate, 2-benzyl-1, 1, 1-trifluoro-3-(quinolin-2-yl)propan-2-ol and trifluoro-3-(isoquinolin-1-yl)-2-(thiophen-2yl)propan-2-ol in a zebrafish embryo model.
