A multimodal (<sup>18</sup>F PET/NIRF) smart optical probe was designed to serve as a molecular imaging tool for noninvasively identifying and grading intracranial astrocytic gliomas as well as for directly visualizing its diffuse infiltrative and deeply-embedded tumor tissue upon application to fluorescence guided surgery. The main issue addressed was developing a strategy that can afford selective detection of intracranial astrocytic gliomas using a single molecular construct that accommodates two different imaging modalities for the purpose of facilitating efficient clinical translation. The design of a multimodal (<sup>18</sup>F PET/NIRF) smart optical probe that affords selective detection of intracranial astrocytic gliomas by reporting on its highly-expressed levels of specific enzyme activity via a molecular logic gate design strategy will be presented.
