Heavy metal ions (HMIs) are toxic contaminants with health on humans at elevated concentrations. Analytical techniques such as flame absorption spectroscopy (FAAS), graphite furnace atomic absorption spectroscopy (GF-AAS), and inductively couple plasma atomic emission spectroscopy (ICP-AES), have been well developed for accurate determination of HMIs concentrations in diverse samples of varying matrix complexities. However, the low volatility of mercury (Hg) and arsenic As, precludes the use of conventional FAAS, GF-AAS, and ICP-AES for Hg and As analyses. These analytical challenges preclude routine analysis of Hg (II) and As (III) in many laboratories. The primary objective of this study is to explore the possible use of fluorescence spectroscopy in conjunction with multivariate partial-least square regression (FS-PLS-1) for a simultaneous and multicomponent analyses of Hg(II) and As(II) ions in human serum albumin samples (HSA). The PLS-1 regression was utilized to correlate changes in emission spectra of Hg (II)-HSA and As (III)-HSA complexes with the Hg(II) or As(III) concentrations in HSA samples. The developed models were later used to predict the concentration of Hg (II) or As (III) in HSA samples. Similar PLS-1 regression study was conducted for the determination of chromium (Cr (VI)) and cadmium (Cd (II)) in HSA samples. The developed PLS-1 regressions resulted in a correlation coefficient (R2) of 0.9808 or better. The capability of the PLS-1 to accurately predict the HMI concentrations in HSA samples was evaluated by a root-mean-square-percent-relative error (RMS%RE) of prediction. In general a RMS%RE ranging between 3.18% and 12.3% was obtained for the prediction of As (III), Cr (VI), Hg (II), and Cd (II) concentrations in HSA samples under physiological condition. A low limit of detection (LOD) of 1.6 x 10-6M for Hg(III), 4.2 x 10-7M for As(III), 1.0 x 10-6M for Cr(VI), and 9.2 x 10-6M for Cd(II), respectively were calculated for the determination of Hg(III), As(III), Cr(VI), and Cd(II) in HSA samples, demonstrating the high sensitivity of the technique.
