Lateral diffusion of membrane proteins is often linked to their cellular signaling and functionality in the cell membrane. Receptor for advanced glycation end-products (RAGE) is one such membrane protein that belongs to immunoglobulin super family and associated with chronic pathological conditions such as cancer, diabetes and inflammation. Even though RAGE signaling has been studied extensively in different disease states, very little is known about RAGE diffusion in the cell membrane. Herein, RAGE lateral diffusion is probed with fluorescence recovery after photobleaching technique. The effect of external stimuli such as cholesterol depletion and F-actin cytoskeletal alteration on the RAGE lateral diffusion and RAGE signaling will be reported. Further, the RAGE downstream signaling is investigated by measuring the phosphorylation of extracellular signal-regulated kinase (ERK) in the native and altered cellular conditions. The correlation between lateral diffusion of RAGE and RAGE signaling will be discussed as well.<br/>
