For more than 3 decades, imino sugar glucosidase inhibitors have been promised as useful antiviral therapeutics. They have been shown to be active against a range of different viral infections, including hepatitis B, C, Dengue, influenza, Ebola and Marburg.  Yet, none have been approved for use in people.  What will it take for an imino sugar glucosidase inhibitor to be become an approved antiviral therapy?  This presentation will provide a brief review of the most promising approaches, taking a candid look at the limitations and failures.  The possibility of identifying non-competitive inhibitors of glucosidases, and progressing either these or traditional imino sugars in combination with other drugs, for management of acute and chronic infections as direct antiviral agents, as well as immunomodulators, will be discussed.  A fresh look at the strengths and weaknesses of this family of drugs and nurture of the non-direct acting antiviral benefits, may be necessary for this perpetual-future class of antivirals to become current.
