Antibody-recruiting small molecules (ARMs) such as dinitrophenyl (DNP) are capable of enhancing the immune response to ARM-modified targets, such as pathogenic bacteria or viruses. Strategies have been developed to deliver ARMs to various pathogens, but to date no strategies have been reported for delivering ARMs to mycobacteria, which include several important human pathogens such as <i>Mycobacterium tuberculosis</i>. Recent work has shown that unnatural trehalose analogues can be metabolically incorporated into the mycobacterial cell wall, providing a possible route for mycobacteria-specific incorporation of DNP or other ARMs. Here, we report the synthesis, characterization, and evaluation of several DNP-modified trehalose analogues.
