Abstract: The enzyme Xanthine Oxidase catalyzes the conversion of hypoxanthine to uric acid. Through this reaction, harmful reactive oxygen species are given off which cause reperfusion injuries, and uric acid can build up causing gout. Caffeic Acid Phenethyl Esters (CAPEs) have been reported to be good antioxidants, and are potential inhibitors for Xanthine Oxidase. Here we describe our study on the synthesis of CAPE derivatives through Malonic Acid Monoester intermediates. Analogues were synthesized to add 1 to 3 hydroxyl groups on the phenyl ring, and to vary the location of the hydroxyl groups around the ring to evaluate what placement and number of hydroxyl group(s) will increase their binding affinity to the enzyme as well as their radical scavenging ability. The biological evaluations are currently ongoing to test their Xanthine Oxidase inhibition and radical scavenger ability.
